This means Spreed WebRTC can’t find static/css/main.min.css.
Did you install Spreed WebRTC from source?
Please post the output of $ find static/css/ from the root directory of spreed-webrtc.

I installed the spreed webrtc server package via your repo https://github.com/strukturag/nextcloud-spreedme#ubuntu-installation-with-packages and downloaded the zipped code (this repo) to /var/www/nextcloud/apps/ and unziped it to a folder called spreedme.
So
root@skynet:/var/www/nextcloud/apps/spreedme# find static/css/
find: ‘static/css/’: No such file or directory
root@skynet:/var/www/nextcloud/apps/spreedme# find extra/static/css/
extra/static/css/
extra/static/css/owncloud.css

I see. Then you need to add this to your webrtc.conf:
[http]
; Full path to directory where to find the server web assets.
root = /usr/share/spreed-webrtc-server/www
Will update the installation instructions to be a bit more clear about this.
