1 - COPS does not write to the calibre database and as far as I know does not lock it either. So probably not a COPS issue!
2 - If calibre cannot write to the (networked) directory/database there is probably a rights issue on your Synology.
Make sure you can write to the networked directory (try to copy a file with windows to the networked directory, and then delete it).

Thanks for the feedback. indeed you are right. COPS had nothing to do with the Calibre/Synology issue.
I also posted my request on https://github.com/SynoCommunity/spksrc/issues/4620
a member of the Synocommunity (hgy59) also kindly answered : "If those few weeks correspond with the DSM update to version 6.2.4 I suppose this is a permission issue.
DSM 6.2.4 introduced some security features, e.g. it warns when you login as admin.
So make sure you do not use the admin user to access the calibre folder on your NAS."
He was spot on ; I used to grant access to the Calibre folder on my NAS via my admin account.
Based on his advice, I created a "Calibre" account, having no admin but R/W access granted to the NAS Calibre folder. Everything went perfectly back as normal immediately.
Thanks for your support and attention anyway, I hope my experience will contribute to others facing potentially the same issue.
