Yes, this was the feature I wanted to implement myself from the very start, yet haven't had time to do so. And I'm a bit away from any development at the moment, so if you can do this, please go on, any additions will be greatly appreciated of course!
My initial idea was to allow to eval certain blocks (like, quoted by backticks or something) and use file local variables substitution there (and probably more, e.g. date time functions or self incrementing counters or wahtever), but your approach may be much easier to actually use, so it sounds good too.

(10 years late) thank you @magnars for adding variables support to restclient.
In the rare cases where the boundaries of a variable are not clear, such as in
:prefix = foo
GET https://example.com/api?search=:prefixbar
I use to do define instead
:{prefix} = foo
GET https://example.com/api?search=:{prefix}bar
I just wonder if you originally had in mind a different approach.
