node-mapnik master requires at least mapnik r2957. I forgot that my binary installer is just a bit older than this, sorry :)
So, you can either use node-mapnik 0.3.x, or get >= r2957 of mapnik installed. I plan to update the binary mapnik installer, but not in the next couple of weeks.

I managed to build node-mapnik from 0.3.1 tag. I'm facing now problems with node.js not finding "mapnik" module running the examples, but I guess it may be something with the paths. I'll give a try again as soon as possible to complete a working installation of node-mapnik.

Ya, you likely need to set:
NODE_PATH=/usr/local/lib/node:$NODE_PATH

@fpedrera - I just posted a new Mapnik OSX installer, cut from Mapnik2 svn r3030, so this should work great with node-mapnik 0.4.0.
