Stop the container, edit wg0.conf and restart. The change will be persistent as long as you don't change one of the server variables like peers, ip, port, etc. which would result in regeneration of wg0.conf
from docker-wireguard.

So you'd just add an IPv6 listening addr to it?

No idea. I don't use or have access to ipv6. You sounded like you knew what you were doing.
